超声辅助离子液体提取艾叶黄酮及其抗氧化、降血糖活性

    摘要:

    以艾叶为研究对象,采用单因素实验及响应面分析法对超声波辅助离子液体提取黄酮进行优化,并对其体外抗氧化和降血糖活性进行测定。结果表明,艾叶黄酮提取最佳工艺参数为:以8 g/L的1-丁基-3-甲基咪唑溴盐([C4mim]Br)作为提取溶剂,超声时间68 min,料液比1:30 g/mL。在此条件下艾叶黄酮得率高达14.36%且高于预测值。体外抗氧化实验发现艾叶黄酮对1,1-二苯基-2-三硝基苯肼自由基(DPPH? )、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS+)和?OH具有较好的清除能力,艾叶黄酮对DPPH? 、ABTS+和?OH的半数抑制浓度(IC50)分别为0.15、0.12和0.6 mg/mL。体外降血糖实验发现艾叶黄酮对α-淀粉酶和α-葡萄糖苷酶的IC50分别为0.59和0.37 mg/mL,表明艾叶黄酮具有良好的抗氧化性和降血糖功效。

    Abstract:

    In this study, the ultrasonic-assisted ionic liquid extraction of Artemisiae argyi leaves flavonoids was optimized using single-factor tests and the response surface method, and its antioxidant and hypoglycemic activities were determined in vitro. The results showed that the optimal process parameters for the extraction of A. argyi leaf flavonoids as follow: 8 g/L 1-Butyl-3-methylimidazolium Bromide ([C4mim]Br) as extraction solvent, ultrasonic time of 68 min, and material-liquid ratio of 1:30 g/mL. Under the optimized conditions, the yield of A. argyi leaf flavonoids was 14.36%. In vitro antioxidant assay showed that A. argyi leaf flavonoids have good scavenging abilities against 1,1-diphenyl-2-trinitrophenylhydrazine radicals (DPPH ?), 2,2-diazo-di (3-ethylbenzothiazole-6-sulfonic acid) diamine salts (ABTS+), and ?OH, with a corresponding half inhibitory concentration (IC50) of 0.15, 0.12 and 0.6 mg/mL, respectively. In vitro hypoglycemic assay showed that A. argyi leaf flavonoids have the inhibitory effect on α-amylase and α-glucosidase, with the IC50 value of 0.59 and 0.37 mg/mL, respectively. The flavonoids of A. argyi leaf had good antioxidant properties and hypoglycemic activity.
