Preliminary Enrichment and Separation of Gallic Acid from Chebulae Fructus by Macroporous Resins

    Abstract:

    The performance and separation capabilities of six resins (NKA-9、AB-8、D-101、HPD-300、HPD-826 and HPD-400) were evaluated to enrich gallic acid in Chebulae Fructus. NKA-9 resin with strong polarity displayed the best performance in these resins due to its good adsorption and desorption capabilities. Subsequently, the static and dynamic adsorption tests of NKA-9 resin for gallic acid in Chebulae Fructus were systematically tested. The optimal parameters were obtained as followed: Adsorption: concentrations of gallic acid in sample solution 0.152 mg/mL; pH 3.0; sample loading amount 3 BV; flow rate 3 BV/h; temperature 25 ?C. Desorption: Distilled water 5 BV, flow rate 3 BV/h, then 30% ethanol 7 BV, flow rate 2 BV/h. The purity of gallic acid in Chebulae Fructus extract increased from 4.7% to 25.3%, and the recovery was as high as 82.6% under above conditions.
