膦酸酯糖苷衍生物的合成与抗肿瘤活性

摘要:以 α-羟基膦酸酯和溴代单糖为中间体,碱性离子液体[Bmim]OH 为催化剂,微波辅助下,以 79.6%~84.1%

的收率合成了系列膦酸酯糖苷化合物(Ⅲa~Ⅲf),经 IR、 HNMR、 CNMR 确认了产物结构。采用溴化噻唑
蓝四氮唑(MTT)法进行了体外抗肿瘤活性测试。结果表明:该类化合物对人胃癌细胞(SGC-7901)和人肺癌
细胞(A549)有增殖抑制作用,其中,含葡萄糖基片段的目标物Ⅲa、Ⅲb 对 SGC-7901 有较优的增殖抑制作用,
IC 50 分别为(12.9±0.9) μmol/L、(14.3±1.1) μmol/L;含半乳糖基片段的目标物Ⅲc、Ⅲd 对 A549 有较显著活
性,IC 50 分别为(15.9±0.9) μmol/L、(14.2±1.3) μmol/L。

Abstract: A series of glycoside derivatives containing phosphonate moiety ( Ⅲ a~f) in yields of
79.6%-84.1% were synthesized by using α-hydroxyphosphonate and bromine substituted monosaccharide
as raw materials, alkaline ionic liquid [Bmim]OH as catalyst under microwave irradiation. Their structures

were confirmed by IR, HNMR and CNMR. These compounds were tested for their in vitro antitumor
activities on some tumor cell lines by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT)
assay. The results indicated that all tested compounds could inhibit the proliferation of SGC-7901 human
gastric cancer cells and A549 human lung cancer cells. Among them, compounds Ⅲa and Ⅲb containing
glucose fragment had better inhibition effect against SGC-7901 with IC 50 values of (12.9±0.9) μmol/L and
(14.3±1.1) μmol/L, respectively. Compounds Ⅲc and Ⅲd containing galactose fragment showed more
remarkable activities against A549 with IC 50 values of (15.9±0.9) μmol/L and (14.2±1.3) μmol/L,
respectively.
