                              基于烯胺酮中间体“一锅两步”法

                 摘要:以苯乙酮为起始原料,  采用“一锅两步”法制备了 β-羰基砜衍生物。首先,在 100  ℃下将苯乙酮衍生物
                 与 N,N-二甲基甲酰胺二甲基缩醛(DMF-DMA)反应生成活泼的烯胺酮中间体;随后,在 80  ℃下加入甲苯为
                 溶剂、以六水三氯化铁/叔丁基过氧化氢为催化氧化体系、磺酰肼为磺酰基来源,经历自由基加成、氧化、亲核
                 取代反应,最终脱去 N,N-二甲基甲酰胺得到终产物,以 30%~64%的收率合成了 18 种具有不同取代基的 β-羰基

                 砜衍生物,采用 HNMR、 CNMR 对终产物进行了结构鉴定。结果表明:该法具有良好的底物普适性,多种取
                 代基团,如甲基、甲氧基、卤素、噻吩、呋喃和萘等都能顺利地发生转化得到相应的 β-羰基砜衍生物。

                 Abstract:  Different  β-keto-sulfone  derivatives  were  obtained  via  a  “one-pot,  two-steps”  method  with
                 acetophenone as starting material. Firstly, acetophenone derivatives reacted with N, N-dimethyl formamide
                 dimethyl  acetal  (DMF-DMA)  to  form  active  enaminone  intermediate  at  100  ℃.  Subsequently,  the
                 intermediate  underwent  radical  addition,  oxidation,  nucleophilic  substitution  at  80  ℃  using  toluene  as
                 solvent and sulfonyl hydrazine as sulfonyl source in the presence of a catalytic amount of ferric chloride
                 hexahydrate/tert-butyl  hydrogen  peroxide.  Finally,  the  final  product  was  obtained  by  removing
                 N,N-dimethyl formamide, eighteen kinds of β-keto-sulfone derivatives were obtained in 30%~64% yields.

                 Their structures were determined by  HNMR and  CNMR. The results show that this method has a good
                 universality of substrates, and various substituents, such as methyl, methoxy, halogen, thiophene, furan and
                 naphthalene can be successfully converted to corresponding β-keto-sulfone derivatives.
