恩施地区野棉花化学成分及其抗肿瘤活性

摘要:采用硅胶、Sephadex LH-20、制备液相等色谱方法对恩施地区银莲花属植物野棉花的乙醇(体积分数为
70%)提取液进行分离纯化,运用 NMR、MS 等方法进行结构鉴定,并考察了化合物的抗肿瘤活性。结果显示,
得到了 10 个化合物,分别为络石苷元(Ⅰ)、牛蒡子苷元(Ⅱ)、绿原酸(Ⅲ)、紫云英苷(Ⅳ)、异槲皮素(Ⅴ)、银椴
苷(Ⅵ)、络石苷(Ⅶ)、牛蒡子苷(Ⅷ)、水杨苷(Ⅸ)、薯蓣皂苷(Ⅹ)。其中,化合物Ⅰ~Ⅱ、Ⅵ~Ⅶ、Ⅸ是首次从银莲
花属中分离得到,化合物Ⅰ~Ⅶ、Ⅸ、Ⅹ是首次从野棉花中分离得到。MTT(四唑盐比色法)测试结果显示,化合
物Ⅱ、Ⅷ、Ⅸ、Ⅹ在 100 μmol/L 时对宫颈癌细胞(HeLa)的抑制率分别为 10.5% 、7.0%、0.5%及 88.0%,其
中化合物Ⅹ的活性最好,高于阳性对照品顺铂的抑制率(80.5%)。

Chemical constituents and antitumor activity of Amenone
vitifolia in Enshi area

Abstract: Extracts with volume fraction of 70% ethanol of Anemone vitifolia in Enshi area of Hubei
province were isolated and purified by column chromatography on silica gel, Sephadex LH-20 and
preparative liquid chromatography. The structures of compounds were identified by NMR, MS and other
methods. Their antitumor activity was tested by MTT assay. The results showed that ten compounds,
including namely, constituents main the lignin, flavonoids and saponins, were identified with trachelogenin
(Ⅰ), arctigenin (Ⅱ), chlorogenic acid (Ⅲ), astragalin (Ⅳ), isoquercitrin (Ⅴ), tiliroside (Ⅵ), tracheloside
(Ⅶ), arctiin (Ⅷ), salicin (Ⅸ) and dioscin (Ⅹ). Compounds Ⅰ ~ Ⅱ, Ⅵ ~ Ⅶ and IX were isolated from
anemone for the first time. Compounds Ⅰ~Ⅶ, Ⅸ and Ⅹ were isolated from Amenone vitifolia for the
first time. MTT test results showed that compounds Ⅱ, Ⅷ, Ⅸ and Ⅹ inhibited the proliferation of HeLa
cells with inhibition rates of 10.5%, 7.0%, 0.5% and 88.0% at 100 μmol/L. Among them, compound Ⅹ
exhibited the best inhibition rate, higher than that of the positive control cisplatin (80.5%).
