菜芙蓉多糖的表征与生物活性

摘要:以菜芙蓉花醇提后的残留物为原料,通过水提醇沉法提取菜芙蓉多糖,测定多糖总糖含量及得率。经
DEAE-52 柱层析分离纯化菜芙蓉多糖〔Abelmoschus manihot (L.) Medic polysaccharide, AMP〕,确定 AMP 单糖
组成,考察 AMP 的体外抗氧化能力,并对 AMP 保护质粒 DNA 及抑制肝癌细胞 HepG2 的增殖作用进行了考察。
结果表明,菜芙蓉花多糖得率为 13.47%,总糖含量为 97.43%,AMP 主要由鼠李糖、葡萄糖、阿拉伯糖、岩藻

糖组成,其物质的量比为 1.44∶5.44∶1.00∶5.37,体外清除自由基能力表现顺序为•O 2 > ABTS • > DPPH• >•OH
> NO•,8 g/L AMP 抑制 H 2 O 2 诱导的 DNA 氧化损伤效果最好,在 0.1250~5.000 g/L 范围内,随着 AMP 质量浓
度的增加 HepG2 细胞的增殖抑制率逐渐增大,说明 AMP 具有抗肿瘤活性。

Characterization and bioactivity of polysaccharide from
Abelmoschus manihot (L.) Medic

Abstract: Polysaccharide was extracted from the ethanol-extracted debris of Abelmoschus manihot (L.)
Medic flowers by water extraction and alcohol precipitation method. The total sugar content and extraction
rate of polysaccharide were determined. Abelmoschus manihot (L.) Medic polysaccharide (AMP) was
separated and purified by DEAE-52 column chromatography and the composition of monosaccharide of
AMP was determined. The bioactivities of AMP, including antioxidant activity in vitro, DNA protecting
activity and hepatic cancer cell HepG2 growth inhibitory effect were investigated. The results showed that
the extraction rate reached 13.47% and the content of total sugar was 97.43%. AMP was mainly composed
of rhamnose, glucose, arabinose and fucose at a molar ratio of 1.44∶5.44∶1.00∶5.37. The order of

radicals scavenging capacities of AMP was •O 2 > ABTS • > DPPH• > •OH > NO•, suggesting AMP had
certain antioxidant activity in vitro. AMP (8 g/L) had the best inhibition effect on inhibiting DNA oxidative
damage induced by H 2O 2. In the range of 0.1250 to 5.000 g/L, with the increase of AMP mass concentration,
the inhibitory rate of HepG2 cells proliferation gradually increased, indicating that AMP had potential
antitumor activity.

菜芙蓉〔Abelmoschus manihot (L.) Medic〕又名 根、茎、叶、花、籽均可利用,含有丰富的总黄酮、

黄蜀葵、金花葵,为锦葵科秋葵属植物,一年生或 多糖、不饱和脂肪酸和微量元素硒、锌等成分 。菜
多年生粗壮直立草本。菜芙蓉为药食两用资源,其 芙蓉花中黄酮类物质含量高(约为花干重的 6%)且
