                    汉麻叶中大麻二酚的分离纯化及其抗抑郁功能

                 摘要:采用柱层析技术对汉麻叶中大麻二酚(CBD)进行分离纯化,并探究其在抑郁症状方面的应用。汉麻叶
                 经预处理后通过超临界 CO 2 萃取得到汉麻浸膏,然后采用树脂两步色谱方法,得到高纯度 CBD,并建立慢性不
                 可预测的轻度应激模型小鼠,采用灌胃 CBD 方式来探究其抑郁行为及对炎症因子的影响。结果表明,通过该法
                 分离纯化提取 CBD 的纯度高达 99.19%;通过动物实验得出,与模型组相比,阳性对照组及 CBD 高低剂量组均
                 显著改善模型小鼠的抑郁行为(P<0.05);模型组小鼠的皮层和海马组织中肿瘤坏死因子(TNF-α)、白细胞介素-1
                 (IL-1β)和一氧化氮合酶(iNos)mRNA 表达量显著升高(P<0.05);CBD 剂量组小鼠的皮层和海马组织中 TNF-α、
                 IL-1β 和 iNos mRNA 表达量均显著降低(P<0.05),说明 CBD 能够有效改善小鼠的抑郁行为和抑制脑组织皮层
                 和海马组织的炎症反应。

                            Separation, purification and antidepressant function of
                                     cannabidiol from Cannabis sativa leaves

                 Abstract: Cannabidiol (CBD)  was isolated and  purified from  Cannabis sativa leaves  by column
                 chromatography. And its application in depressive symptoms was explored. The Cannabis sativa extract
                 was obtained by supercritical CO 2 extraction of pretreated Cannabis sativa leaves. Then, cannabidio with
                 high purity was achieved by a two-step chromatography process. Chronic unpredictable mild stress model
                 mice were established. Intragastric administration of CBD was used to investigate the depressive behavior
                 and the effect on inflammatory  factors. The  results showed that the  purity of CBD extracted  by the
                 above-mentioned method was 99.19%. The depressive behavior of model mice was significantly improved
                 by CBD dose (P<0.05), and the expression levels of tumor necrosis factor (TNF-α), interleukins-1 (IL-1β)
                 and nitric oxide synthase (iNos) mRNA in cortex and hippocampus of model mice were all significantly
                 increased (P<0.05), while the expression levels  of TNF-α, IL-1β and iNos mRNA in the cortex and
                 hippocampus of mice in the CBD dose group were all significantly reduced (P<0.05), indicating that CBD
                 could effectively improve the depressive behavior of mice and inhibit the inflammatory response in the
                 cortex and hippocampus of brain tissue.
