摘要:通过共沉淀法和化学键偶联技术制备出肝癌靶向的 pH 敏感的装载 Ag 2 S 的聚乳酸-羟基乙酸
(SP94-PLGA@Ag 2 S)纳米粒子,将其用于近红外(NIR)Ⅱ区荧光引导的肝癌手术切除。体内外实验结果表
明,在酸性条件下(pH 6.5),SP94-PLGA@Ag 2 S 纳米粒子会发生降解,导致 Ag 2 S 纳米粒子间距增大,荧光恢
复(荧光强度百分比从 20%恢复到 100%),能够实现肝癌组织与正常组织的精准区分。此外,注射
SP94-PLGA@Ag 2 S 纳米粒子后,在近红外Ⅱ区荧光手术导航设备的辅助下,可实现肝癌的有效切除,大大延长
了荷瘤小鼠的生存周期(术后 60 d 生存率由传统手术组的 40%提高到了 90%),可有效降低术后复发率。

Abstract: Hepatocellular carcinoma targeted and pH-sensitive Ag 2S loaded poly(lactic-co-glycolic acid
SP94-PLGA@Ag 2S nanoparticles were prepared by coprecipitation and chemical bond combination
method for near-infrared region Ⅱ fluorescence-guided surgical resection of hepatocellular carcinoma.
The in vitro and in vivo experimental results showed that SP94-PLGA@Ag 2S nanoparticles tended to
produce degradation under acidic condition (pH 6.5), leading to the increase of the distance between Ag 2S
nanoparticles and the recovery of fluorescence (the percentage of fluorescence intensity was restored from
20% to 100%). The degradation of SP94-PLGA@Ag 2S nanoparticles could realize the accurate
differentiation between hepatocellular carcinoma tissues and normal tissues. In addition, the injection of
SP94-PLGA@Ag 2S nanoparticles with the assistance of near-infrared region (NIR) Ⅱ fluorescent surgical
navigation equipment could effectively remove hepatocellular carcinoma, which greatly prolonged the
lifetime (compared with that of the traditional surgery, the survival rate at 60 days after surgery increased
from 40% to 90%) and effectively reduced the postoperative recurrence rate.
