海参斑软骨多糖的纯化、结构及性质分析

摘要:以海参斑软骨为原料,采用水提醇沉法得到粗多糖,通过 Sevag 法除去蛋白,采用 DEAE-52 纤维素和葡
聚糖 G-75 纯化得到组分均一的 3 种多糖 FCPS-1、FCPS-2、FCPS-3。对 3 种多糖进行了化学成分分析及紫外波
长扫描,使用天青 A 比色法对 3 种多糖进行了肝素效价测定,利用 FTIR 和 NMR 对 FCPS-3 进行了结构表征,
采用 GPC 测定了 FCPS-3 的相对分子质量并测定其体外抗氧化活性。结果显示:3 种多糖总糖质量分数均超过
90%,蛋白质量分数低于 3%。紫外波长扫描显示,3 种多糖在 260 nm 以及 280 nm 处紫外吸收峰较弱,表明 3
种多糖中蛋白质及核酸含量较低。天青 A 显色结果显示,FCPS-3 肝素效价最高,达到 1.365 kU/g;红外及核磁
波谱显示,FCPS-3 为酸性多糖,且含有硫酸根,为 β 型吡喃糖。GPC 测定 FCPS-3 的 M w 为 175700 Da,Z 均相
对分子质量为 253032 Da,M n 为 121885 Da。当 FCPS-3 质量浓度为 5 g/L 时,其对 DPPH、ABTS+以及 OH
的清除率分别为 78.63%、96.26%、93.15%。

Abstract: Crude polysaccharides of lumpfish cartilage were obtained by water extraction and alcohol
precipitation, and then Sevag method was used for deproteinization. The purification of crude
polysaccharides was carried by DEAE-52 cellulose and Sephadex G-75, and three kinds of polysaccharides,
FCPS-1, FCPS-2 and FCPS-3 were obtained. These three polysaccharides composition were analyzed and
characterized by UV. Heparin titer of three polysaccharides was measured by azure A colorimetric method.
The structure of FCPS-3 was characterized by FTIR and nuclear magnetic resonance (NMR) spectroscopy.
Moreover, the molecular weight of FCPS-3 was determined by GPC and its antioxidant activity was
measured in vitro. The results showed that the mass fraction of three polysaccharides all exceeded 90%, the
mass fraction of protein was less than 3%. The UV absorption spectra showed that two weak characteristic
absorption peaks of three polysaccharides were observed at 260 nm and 280 nm, indicating that the content
of protein and nucleic acids in three polysaccharides was very low. The heparin titer of FCPS-3 was the
