蓝莓果渣花色苷提取工艺优化及其
提取物的抗肿瘤活性

摘要:在单因素实验基础上,通过正交实验优化蓝莓果渣花色苷在浸提(OSE)、微波辅助(MAE)和超声辅助
(UAE)提取 3 种提取方式下的工艺条件,并对比花色苷组分及其提取物抗肿瘤活性。结果表明:微波辅助提
取法获得花色苷含量优于浸提法和超声辅助提取法,花色苷提取条件为:微波功率 300 W、微波时间 60 s、料
液比为 1∶40 g/mL,在此条件下,花色苷含量 46.880.63 mg C3G/g,3 种提取法均获得 6 种花色苷组分,分别
为飞燕草素-3-葡萄糖苷、矢车菊素-3-葡萄糖苷、矢车菊素-3-芸香糖苷、牵牛花素-3-葡萄糖苷、锦葵素-3-半乳
糖苷和芍药素-3,5-二己糖苷。3 种提取方式获得的花色苷提取物对 HepG2 肝癌细胞和 A549 肺癌细胞生长和侵
染均有抑制作用,且对 A549 肺癌细胞抑制效应更强。

Optimization of Extraction Process of Anthocyanins from
Blueberry Pomace and Its Antitumor Activity

Abstract: Three extraction methods of organic solvent extraction (OSE), microwave-assisted extraction
(MAE) and ultrasonic-assisted extraction (UAE) of anthocyanins from blueberry pomace (BP) were
optimized by single factor test and orthogonal test, and anthocyanins components and their anti-tumor
activity were comparative analyzed. The results showed that MAE was superior to OSE and UAE in
obtaining anthocyanins content. The optimum extracting parameters for anthocyanins from BP were
obtained under microwave power 300 W, extraction time 60 s and solid-to-liquid ratio 1∶40 g/mL. Under
these conditions, anthocyanins contents of 46.88±0.63 mg C3G/g were achieved. The three extraction
methods all obtained six anthocyanins components, which were delphinidin-3-glucoside, cyanidin-3-glucoside,
cyanidin-3-rutinoside, petunidin-3-glucoside, malvidin-3-galactoside and peonidin-3,5-dihexoside. Anthocyanins
obtained by three extraction methods inhibited the growth and invasion ability of HepG2 hepatoma cells
and A549 lung cancer cells, and had stronger inhibitory effect on A549 lung cancer cells.
