                              响应面法优化提取黄檗果实总酚和


                                        总黄酮及其抗氧化活性

                 摘要:在单因素实验基础上,根据 Box-Behnken 实验设计对提取条件进行分析和优化,以考察各因素对黄檗果
                 实总酚(TP)和总黄酮(TF)超声辅助提取率的影响,并进一步研究 TP 和 TF 与体外抗氧化活性〔DPPH•清除
                 能力、总自由基清除能力(TRAP)、铁离子还原能力(FRAP)〕的相关性。结果表明,黄檗果实 TP 和 TF 最佳
                 超声辅助提取工艺条件为:乙醇体积分数 43%,提取温度 70  ℃,pH 值 11,超声功率 500 W,液固比 30∶1(mL/g),
                 超声时间 40 min,TP(以每克干样品中酚酸类化合物相当于没食子酸 GA 的质量表示)和 TF(以每克干样品中
                 黄酮类化合物相当于芦丁 RT 的质量表示)得率分别为(7.09±0.38)mg  GA/g 和(20.41±0.47)mg  RT/g;相关
                 性分析结果表明,TP 和 TF 含量与三种体外抗氧化活性检测结果均为极显著相关(P<0.01),说明 TP 和 TF 可
                 能是黄檗果实提取物抗氧化活性的物质基础。

                   Optimization of extraction of total phenolics and total flavonoids from

                               Phellodendron amurense fruit by response surface

                 Abstract:  On  the  basis  of  single-factor  test,  the  extraction  conditions  of  total  phenolics  (TP)  and  total
                 flavonoids  (TF)  from  Phellodendron amurense  fruit  were  optimized  by  Box-Behnken  design.  The
                 antioxidant activities in vitro of the extracts were evaluated by studying 1-diphenyl-2-picrylhydrazyl free
                 radical (DPPH•) scavenging assay, total radical antioxidant power (TRAP) and ferric reducing antioxidant
                 (FRAP).  Their  correlation  between  antioxidant  activities  of  TP  and  TF  and  their  contents  were  further
                 investigated. The results showed that the optimal extraction conditions of TP and TF from Phellodendron
                 amurense fruit were as follows: ethanol volume fraction 43%, extraction temperature 72  ℃, pH value 11,
                 ultrasonic power 500 W, liquid/material ratio 30∶1 (mL/g), and extraction time 40 min. The yield of TP
                 (expressed as equivalent to gallic acid (GA) per gram of dry sample) and TF (expressed as equivalent to
                 rutin (RT) per gram of dry sample) were (7.09±0.38) mg GA/g and (20.41±0.47) mg RT/g under the above-
                 mentioned  conditions.  The  correlation  analysis  results  showed  that  the  contents  of  TP  and  TF  were
